Drug intelligence / Profile preview
[89Zr]Zr-DFO-H4-Fc is a radiolabeled immuno-PET imaging agent developed by the University of Wisconsin-Madison. It consists of H4-Fc, a nurse shark-derived variable new antigen receptor (VNAR) single-domain antibody fused to a human IgG1 Fc scaffold, which is site-specifically conjugated to a deferoxamine (DFO) chelator and radiolabeled with the positron-emitting isotope zirconium-89 ([89Zr]Zr). The agent specifically targets fibroblast activation protein alpha (FAP), a cell-surface serine protease highly expressed on activated fibroblasts at sites of tissue remodeling, chronic inflammation, peripheral nerve injury, and in the tumor microenvironment of solid tumors. [89Zr]Zr-DFO-H4-Fc is being investigated in preclinical studies as a diagnostic tool to non-invasively detect and localize chronic neuropathic pain and neuroinflammation, as well as for potential theranostic applications in oncology.
